Small Molecule Targeting of PAI-1 Function: A New Therapeutic Approach for Treatment of Vascular Stenosis
نویسندگان
چکیده
Plasminogen activator inhibitor-1 (PAI-1; SERPINE1) is a clade E1 member of the serine protease inhibitor (SERPIN) superfamily and the major physiologic inhibitor of the urokinase (uPA) and tissuetype (tPA) plasminogen activators. Elevated PAI-1 expression is a significant causative factor in vascular disease and a major contributor to the pathophysiology of diabetes, metabolic syndrome, stroke, atherosclerosis and restenosis, particularly in the setting of increased vessel TGF-β1 [1-3].

A small molecule inhibitor to plasminogen activator inhibitor 1 inhibits macrophage migration.
OBJECTIVE Macrophage (Mϕ) migration rests on the adhesion/detachment between Mϕ surface components and extracellular matrixes, and the contribution of numerous inflammatory disorders. Plasminogen activator inhibitor (PAI)-1, a serine protease inhibitor, influences Mϕ motility through an action distinct from its classical modulation of the plasmin-based fibrinolytic process. We rely here on a sm...
متن کاملTargeting plasminogen activator inhibitor-1 inhibits angiogenesis and tumor growth in a human cancer xenograft model.
Cancers of the urinary bladder result in aggressive and highly angiogenic tumors for which standard treatments have only limited success. Patients with advanced disease have a 5-year survival rate of less than 20%, and no new anticancer agent has been successfully introduced into the clinic armamentarium for the treatment of bladder cancer in more than 20 years. Investigations have identified p...
متن کاملالقای آپوپتوز وابسته به p53 در ردهی سلولی لوسمی لنفوبلاستیک حاد پیشساز لنفوسیت B (NALM-6) توسط مولکول کوچک RITA
Background and Objective: The use of low-molecular-weight, nonpeptidic molecules that degrade the interaction between the p53 protein and its negative regulator MDM2 (Murine- double minute colon 2) is a new therapeutic strategy for treatment of various types of cancer. One of these agents is RITA (reactivation of p53 and induction of tumor cell apoptosis) which binds to p53 protein and inhibits...
متن کاملThe effect of microRNA-125 on the adhesion molecule expression of integrin beta2 and adhesive determination of endothelial cells isolated from human aorta to monocyte
Background: The immune-mediated responses in vascular cells may include the increased expression of endothelial adhesion molecules, leukocyte rolling and infiltration, cellular lipid dysregulation and vascular smooth muscle cells (VSMCs) differentiation. Investigating the cellular and molecular events involved in the rolling process is useful for treatment or prevention of the vessel stenosis es...
متن کاملP122: Small Molecules as Chemical and Pharmacological Tools for Neuroinflammatory Diseases Treatment (with Emphasis on Multiple Sclerosis)
Multiple Sclerosis (MS) is a neuroinflammatory disease resulting in degeneration of the myelin sheaths and death of oligodendrocytes. So far, several strategies have been introduced to control the disease. Treatment with small molecules is one of the strategies that have recently attracted the attention in the scientific community. These molecules that target epigenetic and other cellular proce...
